| I. Background | |
| APOE 是一種膽固醇載體，參與腦內(nèi)脂質(zhì)運(yùn)輸和修復(fù)。APOE 是一種多態(tài)蛋白，具有 APOE4、APOE3 和 APOE2 三種異構(gòu)體，它們之間存在兩個氨基酸替換（分別為 Arg/Arg、 Cys/Arg 和 Cys/Cys），導(dǎo)致三級結(jié)構(gòu)不同，并可能改變 APOE 的功能。盡管異構(gòu)體之間存 在相似性，但它們在發(fā)生 AD 的風(fēng)險(xiǎn)方面存在重大差異，其中 APOE4 是 AD 發(fā)生的強(qiáng)烈危 險(xiǎn)因素。APOE3 是最常見的亞型，不影響患 AD 的風(fēng)險(xiǎn)，而 APOE2 與患 AD 的風(fēng)險(xiǎn)降低有關(guān)。 | |
