| 補體 C5a 是由補體系統(tǒng)經(jīng) C5 轉(zhuǎn)化酶裂解 C5 產(chǎn)生的強效炎癥介質(zhì),作為核心過敏毒素 通過結(jié)合受體 C5aR1/C5aR2 驅(qū)動中性粒細胞趨化、氧化爆發(fā)及炎性因子(IL-6/TNF-α)釋 放,在膿毒癥中誘發(fā)“細胞因子風暴”,在自身免疫?。ㄈ珙愶L濕關(guān)節(jié)炎)中促進組織侵 蝕,在腫瘤微環(huán)境募集髓系抑制細胞(MDSC)介導免疫逃逸。 C5aR1(CD88)是補體過敏毒素 C5a 的高親和力 GPCR,主要表達于髓系細胞(中性 粒細胞、單核細胞、肥大細胞),通過激活 Gi/Gq 蛋白-PI3K/MAPK 通路驅(qū)動炎癥級聯(lián)反 應。 |
